Be sure that pop3 protocol is switched ON for thiw email account before test, use port 995 and ssl and read this for configuration:

Another question:

What does? Post replay or new topic: No Yes

... to Kunena mean?

Does "No" mean NO posting to Kunena?
What does "replay" mean? Do you mean "reply"?

I only want to reply to existings Kunena posts.

1. I am using post_by_email_pro_j15_1.5.0 for J1.5.26. The admin page is different. Is something wrong? Please see below.

2. Please confirm that when set to YES TO REPLY, the post will NOT automatically be posted in the original kunena category? Reply posts will only be posted to "one" particular category? not necessarily the original category?

No. Joomla 1.5 and Joomla 2.5 versions of Post by Email Pro are with different look.
